' Meanwhile, tolerance of doses near 1 g (or up to 3 g) of daily intake of NAM, even during long-term administration, has been demonstrated in many studies. Examples are the European Nicotinamide Diabetes Intervention Trial (ENDIT), where children were treated with 25â50 mg/kg a day for 5 years without reported adverse effects [20, 97] and a recent clinical study which showed that intake of 1.5 g NAM twice daily for 6 months was safe in elderlies [NCT00580931]. These studies assure safety in the current wide-spread practice of long-term dietary intake of 500â1000 mg per day. Although headache, dizziness, and vomiting occurred in healthy humans when NAM at doses up to 6 g was ingested on an empty stomach [43], these adverse effects were minor and resolved upon termination.
Note that problems tended to start to occur in the area of 6 grams on an empty stomach. That is a huge dose for most supplements and I haven't seen studies suggesting that such high doses are useful and healthful.
In previous testing I have found Nicotinamide or Niacinamide to be very tolerable and noticed no apparent side effects at the dosages I tested. This is a useful addition to EC's list of glaucoma remedies.

Hi NK,To be clear on dosing, none of the referenced studies used nicotinamide, niacinamide or nicotinic acid(Niacin) directly in the eyes, but rather used human oral dosing or equivalent animal model dosing of Niacinamide in the 3 gram day dosing area for humans taken orally. This oral dosing area has been shown to be safe in previous studies.
The 1 hour of tingling and burning feeling you experienced does not sound safe in my opinion and is not reflective of the oral dosing used in the multiple studies which is known to be safe.
